Synthesis, Characterization, and Anti-Tumor Activity of Isatin Derivatives: Insights from Spectroscopic and Biological Studies

Indole alkaloids, particularly isatin and its derivatives, exhibit diverse pharmacological activities, including antibacterial, antiviral, anti-inflammatory, and anti-cancer effects. This study focuses on the synthesis, characterization, and biological evaluation of novel isatin derivatives as potential anti-cancer agents. Structural characterization was conducted using infrared (IR) spectroscopy, proton and carbon nuclear magnetic resonance (1H NMR, 13C NMR), and elemental (CHN) analysis, providing insights into molecular modifications. Biological evaluation using normal fibroblast and lung cancer cell lines demonstrated that the compounds induced apoptosis and arrested the cell cycle in cancer cells, indicating promising anti-tumor activity. Future research at the Hiroshima Research Institute for Synchrotron Radiation Science (HiSOR) will utilize circular dichroism (CD) spectroscopy to study protein conformations. Synchrotron-based CD offers high sensitivity and resolution, enabling detailed analysis of protein folding, stability, and ligand interactions. These findings will aid in optimizing drug design and understanding biomolecular mechanisms, supporting the development of novel therapeutic strategies.
